MECHANISM OF ACTION

Ondansetron works by blocking the action of serotonin, a natural substance that may cause nausea and vomiting. Ondansetron is a selective antagonist of the serotonin receptor subtype, 5-HT3. Cytotoxic chemotherapy is associated with the release of serotonin from enterochromaffin cells of the small intestine.

Presumably initiating vomiting reflux through stimulation of 5-HT3  receptor located on vagal afferents. Ondansetron may block the initiation of this reflux. Ondansetron and the other members of the strong drug family are serotonin antagonists. . Which means these drug blocks the serotonin receptor in the chemoreceptor trigger zone ( CTZ) within the medulla oblongata.

The chemoreceptor trigger zone ( CTZ)  communicates with the vomiting center to initiate vomiting. By blocking the serotonin receptor, there`s less serotonin that enters the chemoreceptor trigger zone (CTZ),  which decreases communication with the vomiting center. Ultimately, the patients experience reduced nausea and vomiting.
